Murashige & Skoog Basal Salts provide a comprehensive nutrient formulation for plant tissue culture. The formulation contains essential macronutrients, micronutrients, vitamins, and glycine to support cultured plant tissues.
The medium provides nutritional support for cellular metabolism, growth, tissue development, and differentiation. Glycine contributes to the nutritional requirements and vigor of cultured plant tissues.

Assay Protocol: Murashige & Skoog Media Preparation
General procedure for preparing 1 liter of tissue culture medium using MS basal salts:
- Add approximately 800 mL of distilled water to a suitable beaker.
- Add 4.54 g of MS medium for 1 liter of final medium.
- Add 30 g of sugar per liter of medium.
- Add PPM at approximately 1–2 mL/L, if required by your validated protocol.
- Bring the total volume to 1000 mL with distilled water.
- Adjust the pH to approximately 5.4–5.8.
- Add a gelling agent if a solid medium is required: 3–4 g/L gellan gum or 6–8 g/L agar.
- Continue stirring until the components are adequately dispersed.
- Autoclave the prepared medium for approximately 20 minutes at 15 psi and 121°C, according to the validated laboratory protocol.
Prepare the medium according to the formulation specified on the product label. It is recommended to use an entire container at once where applicable. Heat-labile substrates should be added after autoclaving using an appropriate sterile technique.
